Abstract

âº Mutations in the ferritin light chain gene make the molecule susceptible to oxidation. âº The oxidation of mutant ferritin causes loss of ferritin structure. âº Iron-catalyzed oxidation of mutant ferritin causes polypeptide cleavage. âº Oxidative modifications may contribute to inclusion body formation and neuropathology. âº Oxidative damage of mutant ferritin is a novel pathogenic mechanism in ferritinopathy.
