Abstract
⺠Tumor cells organize a specialized chaperone network within their mitochondria that consists of at least Hsp90 and TRAP-1. ⺠Shepherdin and Gamitrinibs are compounds that target the mitochondrial Hsp90 chaperone network. ⺠Gamitrinibs cause cell death in cancer cells that depends on the mitochondrial permeability transition pore. ⺠Low dosages of Gamitrinibs cause a stress response, resulting in a sensitization of apoptotic resistant cancer cells to TRAIL. ⺠Gamitrinibs cause energy depletion in tumor cells, mediating inhibition of mTOR - signaling.
