Abstract
► Checkpoint kinases use 14-3-3 proteins to establish and maintain a G2 cell cycle checkpoint by targeting Cdc25 and Wee1. ► Cdk1 and Plk1 inactivate checkpoint kinase pathways targeting Cdc25B and Wee1 binding to 14-3-3 to drive mitotic entry. ► 14-3-3 proteins prevent apoptotis by binding to BH3-containing proteins, FOXO transcription factors and apoptotic kinases. ► JNK directly phosphorylates 14-3-3 proteins to block their anti-apoptotic effects and drive cell death.
