► Hydropericardium syndrome is an important disease its efficacious vaccine is needed. ► Amplification of the capsid protein gene (penton base) of fowl adenovirus-4. ► Cloning and expression of the viral gene (1587 bp) in fusion with His tag. ► Purified penton base protein (65 kDa) conferred 90% protection against viral challenge. ► Penton base is found to be a candidate for subunit vaccine against fowl adenovirus-4.
