Abstract
► Integrase-defective lentiviral vectors (ID-LVs) efficiently infect DC precursors. ► Integration of ID-LVs into the genome is dramatically reduced, enhancing safety. ► ID-LVs co-expressing GM-CSF/IL-4 or GM-CSF/IFN-α induce monocytes to differentiate into DCs. ► Lentivirus-induced DCs (iDCs) can be co-programmed with full length antigens. ► Human iDCs co-expressing a CMV antigen stimulated CD8+ T cells in vitro and in vivo.
