Abstract

With vinyl ester resin as monomer, ethylene glycol dimethacrylate (EDMA) as the crosslinker, dodecanol as porogenic solvents, and 2,2'-azobisisobutyronitrile (AIBN) as initiators, a poly(vinyl ester resin-co-ethylene dimethacrylate) monolithic column was prepared by “in-situ” process. The monolith was further derivative by 3-glycidoxypropyltrimethoxysilane and then hydrolyzed in 0.25 M sulfuric acid to prepare a diol column. The effects of pH value and buffer concentration on the retention of IgG were investigated and the separation of IgG from human plasma was operated. The separation of IgY from chicken egg yolk at different buffer concentration and the effect of spacer arm on the separation were also investigated. The results show that the monolith with spacer arm had a higher separation capacity. Moreover, the adsorption capacity of the column was investigated and the maximum adsorption amount was 35 μg−1.
