Abstract

Three affinity matrices for the adsorption of endotoxin from aqueous solutions were studied by covalently bonding polymyxin B with varying lengths of arm to Sepharose CL-4B. The ligand densities and the endotoxin-adsorption efficiency increased with increasing length of the spacer arm. It was shown that in the case of Matrix 3, when the NaCl concentration ranged from 0.05 to 0.5 M, pH ranged from 6 to 10, temperature ranged from 25 to 55°C; and flow rates ranged from 0.20 to 0.80 ml/min, the adsorption efficiency was higher than 90%.
