Abstract
•Quantifying biopharmaceutical proteins in complex biological matrices by LC-MS/MS.•Enrichment at the peptide level after proteolytic digestion.•Chemical derivatization of peptides may enhance ionization efficiency.•Sample-preparation aspects of quantifying biopharmaceutical proteins.•Automating the entire analytical procedure.
Part I of this review discusses sample-preparation aspects of quantifying biopharmaceutical proteins in complex biological matrices by LC-MS/MS with a focus on blood-derived body fluids. We conclude Part I with a short overview over options for automating the entire analytical procedure, which is indispensable for routine applications in pharmacokinetic and clinical studies.
