► YlLac1p is essential for hyphal growth in Yarrowia lipolytica. ► Y. lipolytica GlcCer consists of a C16:0 fatty acid and a C18:2 long chain base. ► The entire TRAM/Lag1/CLN8 (TLC) domain is crucial for the function of YlLac1p. ► Yllac1Δ neither polarizes lipid rafts nor forms normal vacuoles. ► Ylpex5 mutation partially suppresses the defective hyphal growth of Yllac1Δ.
