•A “green” electrochemical route for the preparation of graphene/Nafion film was proposed.•The ER-GO/Nafion/GCE exhibited high electroactivity to the oxidation of 8-OH-dG.•The ER-GO/Nafion/GCE can effectively decrease the background current of uric acid.•The interference of uric acid on the detection of 8-OH-dG can be completely eliminated by uricase.
Graphene/Nafion nanocomposite film was fabricated on the glassy carbon electrode via a “green” electrochemical reduction route. The X-ray diffraction patterns and UV–Vis spectra confirmed the complete reduction of graphite oxide. The graphene/Nafion nanocomposite film showed high electrochemical performance on the oxidation of 8-hydroxy-2́-deoxyguanosine with a wide linear range (0.07–33.04 μM) and a low detection limit (1.12 nM). Nafion played an important role in suppressing the oxidation current of uric acid, which is a major interferent to the electrochemical detection of 8-hydroxy-2́-deoxyguanosine. The experiment results proved that the interference of uric acid to the determination of 8-hydroxy-2́-deoxyguanosine can be completely eliminated by uricase. The electrochemically reduced graphene/Nafion nanocomposite film modified electrode will have potential application to the determination of 8-hydroxy-2́-deoxyguanosine in real samples.
