Abstract

A method was developed to make a switch between high performance liquid chromatography (HPLC) and ultra performance liquid chromatography (UPLC) for analyzing a complex sample. In the study, the water extract of Radix Salvia miltiorrhizae was taken as a model sample. Retention parameters (a and c values) of 12 peaks were calculated using five linear gradients on both HPLC and UPLC, and the relationship between the a and c values for HPLC and UPLC was developed, respectively. With the linear equation, the a and c values on HPLC and UPLC could be transferred to each other, and prediction and optimization was realized with the calculated a and c values in the CSASS software. A good linear relationship between a and c values for HPLC and UPLC was developed. The R2 for the a and c linear equations were 0.9914 and 0.9936, respectively. The a and c values of the water extract of Ligusticum chuanxiong on UPLC was calculated by this method, and the analysis on UPLC was predicted by the calculated a and c values. The chromatogram on UPLC was similar to the simulant chromatogram. The method switch between HPLC and UPLC allowed compatible data for both techniques. This method was simple and accurate. It facilitated the predication and optimization of separation, for a complex sample.
