Abstract

Oxalate salt of 1-(isopropylamino)-3-(5-((isopropylamino) methyl)-2-methoxyphenoxy) propan-2-ol (PP-24) is a newly synthesized putative β-adrenoreceptor antagonist and recently we have reported β-antagonistic and antihypertensive activity of PP-24 having the propanolamine group. In this investigation, we are reporting cardiovascular system specific pharmacological activity of the PP-24. The objective of the study was to evaluate effectiveness of this compound in cardiovascular disease conditions in laboratory animals. The antihypertensive activity of PP-24 was studied using Sprauge-Dawley rats, in uninephrectomised deoxycorticosterone acetate (DOCA) salt induced model of hypertension. Anti-arrhythmic activity against ventricular arrhythmias and lethality induced by ouabain was evaluated in guinea pig model. While the cardio protective effect of the compound against cardio toxicity as seen in myocardial infarction was studied using isoproterenol induced myocardial necrosis model in albino mice. Significant reduction in mean arterial pressure (MAP) and heart rate was observed in hypertensive rats treated with PP-24 (10 mg/kg). In ouabain induced ventricular arrhythmia and lethality model, results indicated that PP 24 has anti-arrhythmic activity. Pretreatment with PP-24 (10 mg/kg) significantly reduced the activities of the serum marker enzymes CKMB, LDH, and AST and that of total cholesterol. The effectiveness of PP-24 in the treatment of antihypertensive, anti-arrhytmic and cardioprotective activity in laboratory animals may be due to blockade of β-adrenoreceptors.
