Abstract
In this study, α-amylase was immobilized onto chitosan bead via adsorption with glutaraldehyde. The immobilized α-amylase was exposed in 0.15 T static magnetic field for a period of time and characteristics including activity, kinetic parameters, pH, operational and storage stability were investigated. Under the magnetic field, the activity was increased by 19.53%, Km value was decreased from 11 mg/mL to 9 mg/mL and Vmax increased from 0.02 g/min to 0.05 g/min. Operational and pH stability were improved after magnetically exposed treatment, and the immobilized α-amylase was more stable and could be restored for 30 days with 19.4% loss in activity.
