Abstract
Graphical abstractDownload full-size imageThis report focuses on the concept, underlying chemistry, methods and applications of the ligation techniques that are devolved for the assembly of polypeptides and proteins. Ligation methods work on a chemoselective capture step followed by an intramolecular acyl transfer. An elaborated account of native chemical ligation along with selected case studies is furnished. Expressed protein ligation, traceless Staudinger ligation and the OâN acyl migration protocol (click or switch peptide chemistry) are also dealt with.
