Abstract
Short interfering RNAs (siRNAs) containing P3â²âN5â² phosphoramidate linkages were successfully synthesized by introducing 2â²-deoxy-2â²-fluororibonucleoside and 5â²-amino-2â²,5â²-dideoxy-2â²Î±-fluororibonucleoside in succession. It was found that the introduction of 5â²-amino-2â²,5â²-dideoxy-2â²Î±-fluororibonucleosides into siRNAs improved the nuclease-resistant properties of the siRNAs without loss of their silencing efficacy.
