► Helium isotopes tunnel through nanoporous graphene barrier at room temperature. ► Temperature gradient across barrier causes difference in isotope ratios. ► Classical barrier transmission cannot lead to isotopic enrichment. ► Tunneling mass-dependence enriches heavy isotope on cold side. ► Zero-point differences of the transition state enrich heavy isotope on hot side.
