► Short-term exposure of 2-cells to fluoxetine enhances mouse blastocyst formation. ► The enhancive effect of fluoxetine is resulted from CaMKII activation. ► Long-term exposure of 2-cells to fluoxetine decreases mouse blastocyst formation. ► The inhibitory effect of fluoxetine is mediated through TREK channel gating.
