► Aquaporin-8 is expressed in mitochondria of human hepatoma HepG2 cells. ► Aquaporin-8 knockdown impairs mitochondrial H2O2 release and increases ROS. ► Aquaporin-8 knockdown causes ROS-induced mitochondrial depolarization and cell death. ► Mitochondrial permeability transition blockage prevents depolarization and cell death.
