Abstract
► We investigate the role of two different Lrp5 HBM mutations in modulating mechanotransduction. ► The A214V mutation confers increased mechanosensitivity to the periosteal surface of the tibia. ► The G171V mutation lowers the osteogenic threshold on the endocortical surface of the tibia. ► Both gain- and loss-of function mutations in Lrp5 appear to modulate mechanotransduction.
