► Osteopontin is involved in the initiation of immune responses to diverse pathogens. ► Trypanosoma cruzi infection induces osteopontin release from murine macrophages. ► In vivo neutralization of osteopontin augments mice susceptibility to infection. ► In vivo neutralization of osteopontin reduces Th1 and Th17 cytokine production. ► Endogenous osteopontin helps regulate protective Th1 and Th17 responses to T. cruzi.
