► The dorsal premammillary nucleus (PMd) has a role in processing predatory threats. ► NMDA/NO pathway possibly modulate defensive reactions in this structure. ► Intra-PMd injection of the NMDA antagonist or the NO synthase inhibitor attenuated behavioral responses induced by live cat.
