•Lipase molecules were orientated through controlled adsorption kinetics.•The catalytic domain was exposed on hydrophilic silica due to conformation changes.•Hydrophobic surfaces inhibited the opening of lipase’s lid domain.•Lipase activity correlated well with lipase orientation in engineered biomaterials.
