► Magnetization transfer (MT) is sensitive to myelin deficiency in a mutant canine model. ► All parameters of the two-pool MT model are affected by dysmyelination. ► Strongest age- and disease-related changes were observed for the bound pool fraction. ► Traditional MT ratio (MTR) lacks specificity to myelination and dysmyelination.
