► Depression is related to hyperactivation of an amygdala-centered (para)limbic network. ► Duloxetine attenuates affective processing in same neurocircuitry in healthy humans. ► Duloxetine potentiates effective communication between subparts of the network. ► This may represent mechanism through which duloxetine normalizes negativity bias in depression.
