Abstract
⺠Homologous recombination is a safe and effective strategy for gene therapy. ⺠Engineered nucleases increase gene targeting for PID genes in human cells. ⺠Gene targeting can correct mutations or insert transgenes into specific loci. ⺠Nuclease efficacy can be hampered by toxicity and chromatin configuration.
