► β2GPI-dependent aPL induce fetal loss/growth retardation in naïve pregnant mice. ► Histogenesis/organogenesis inhibition and not inflammation is involved in our model. ► The synthetic peptide TIFI competes with β2GPI binding and protects pregnant mice. ► Inhibition of β2GPI tissue expression represents an innovative therapeutic approach.
