► To develop NS3 gene sequencing method for genotyping and PI resistance profiling. ► Validation on 139 plasma samples from patients infected with HCV genotype 1-6. ► Phylogenetic analysis showed concordance of genotypes in NS5b and NS3 genes. ► PI resistance mutations were successful analyzed using Geno2Pheno [hcv] V1.0.
