Abstract
⺠We describe a method for studying the axonal transport of endogenous proteins fused to HaloTag® in rat sympathetic neurons in compartmented cultures. ⺠Applying HaloTag ligand to the distal axons allows the tracking of labeled proteins during retrograde transport. ⺠We demonstrate that the transport of HaloTag protein alone does not interfere with tracking the transport of HaloTag fusion proteins. ⺠We demonstrate the retrograde transport of HaloTag protein fused to the p75 neurotrophin receptor. ⺠We also demonstrate the anterograde transport of HaloTag protein fused to tubulin.
