► Plasmodium yoelii ADA shows higher specificity for 2′deoxyadenosine than adenosine. ► Histidine and thiol groups are involved in enzyme activity. ► EHNA competitively inhibits ADA activity with 0.5 μM Ki. ► Enzyme is a monomer of ∼41 kDa subunit. ► GdmCl induced ADA unfolding follows a biphasic path.
