► Acute impact of antimicrobials affected the stoichiometry of methane generation. ► Acetate was removed but methane generation dropped with increasing antimicrobial dose. ► Antimicrobials prevented complete utilization of acetate in metabolic reactions. ► Substrate binding by uncompetitive inhibitions explains the observed results.
