► A new property of TreS from Thermus thermophilus HB-8 was found and described. ► Enzyme protein can act on sucrose and catalyze trehalulose formation. ► A simple, fast, and efficient process on producing and purifying trehalulose. ► Other sugars were removed, while trehalulose was preserved by S. cerevisiae cells.
