► Brief, transient expression of RCAN1 proteins promotes oxidative-stress adaptation. ► Chronic RCAN1-1L overexpression can promote neurodegeneration and neuronal death. ► RCAN1-1L-induced tau hyperphosphorylation may stimulate neurodegeneration. ► RCAN1-1L-induced mitochondrial dysfunction may stimulate neurodegeneration. ► Animals chronically overexpressing RCAN1-1L may be useful Alzheimer models.
