► Mice differing in life span-extending response to caloric restriction (CR) were studied. ► Age-related increases in protein oxidation were dependent on genotype and tissue. ► Attenuation of protein oxidation by CR was genotype and tissue specific. ► The effect of CR on protein oxidation did not fully accord with life-span extension.
