► C-Myc activation affects myocardial substrate utilization for the TCA cycle. ► Fatty acid oxidation increases with established compensated cardiac hypertrophy. ► Substrate oxidation is not altered prior to the development of hypertrophy. ► Myocardial O-GlcNAcylation was increased by c-Myc.
