Abstract
⺠MC5R internalizes independently of β-arrestins and recycles to cell surface. ⺠α-MSH binding to MC5R promotes a biphasic ERK activation, an early and a late phase. ⺠The early peak of pERK is Gi-driven; the sustained is β-arrestins dependent. ⺠A small fraction of pERK1/2 translocates to the nucleus inducing c-Fos expression. ⺠ERK lead to p90RSK and MSK1 phosphorylation in Hela cells and 3T3-L1 adipocytes.
