La farmacocinética/farmacodinámica de los iPDE5. ¿Clínicamente significativos? La farmacocinética clínica de los inhibidores de la fosfodiesterasa tipo 5 en disfunción eréctil

Differences in the clinical pharmacology of the three PDE5 inhibitors, sildenafil, vardenafil and tadalafil, are determined not only by the pharmacokinetic characteristics of these drugs, but also by their activity profile.This review presents in a comparative way the main characteristics of the pharmacokinetic profile of the three PDE5 inhibitors, including their bioavility and absortion rate, biopharmaceutical classification, elimination mechanisms and metabolic profile including active metabolites and the potential interactions between drugs under physiologic and pathophysiologic conditions. The aim of this review is provide clinical pharmacologic data to allow the clinician the use of these drugs from a rational scientific aproach to the treatment of erectile dysfunction.
