► CanPIs structure is predominantly β-sheet and unordered. ► CanPI-7 also reveals signature of polyproline fold in CD spectra. ► Inhibition kinetics reveals higher potency of multidomain CanPIs against proteases. ► CanPIs highly stabile under extreme pH, temperature and denaturing conditions. ► Structure prediction, MALDI-TOF-MS and docking reveal binding mechanism of multidomain CanPI-7.
