► Actin-binding domains of vinculin and metavinculin differ in thermal stability. ► Cardiomyopathy-related mutation increases metavinculin residency in cell adhesions. ► Presence of metavinculin in costameres is strictly muscle fiber-type dependent. ► Metavinculin decorates costameres of fast glycolytic but not slow oxidative fibers. ► Metavinculin expression in skeletal muscle correlates with mechanical shear stress.
