► We identify the PHP domain as a novel target of antibiotics. ► DNA polymerase PolC has activity against pNP-TMP. ► FQE inhibits CpsB & PolCPHP activity. ► The targeting of PolCPHP may be responsible for FQE growth inhibitory effects.
