► AMPK phosphorylates VASP at a novel site, serine 322, and not at known PKA sites. ► Phosphorylation prevents VASP binding to actin filaments. ► Inhibition of AMPK phosphorylation results in VASP accumulation at cell-cell contacts. ► Inhibition of AMPK phosphorylation results in increased cell-cell adhesion.
