Abstract
► The expression of phage-displayed disulfide-stabilized sc-dsFv libraries can be drastically enabled with novel signal sequences. ► The effective signal sequence variants in the h-region are not necessarily composed of consecutive hydrophobic residues. ► Synthetic sc-dsFv library complexities are comparable with the scFv-based libraries. ► The disulfide-stabilized sc-dsFv format is a viable alternative for scFv-based applications.
