Abstract
⺠Most pathogenic mutations possess strong directional selection, i.e., enhancing hydrophobicity or decreasing negative and increasing positive charge. ⺠Mutation-induced changes may strengthen the interactions between PrP and facilitating factors. ⺠The findings also have significant implications for exploring potential regions involved in the conformational transition from PrPC to PrPSc.
