Abstract
⺠A novel GLP-1 gene therapy utilizing a strategy involving site-specific genome integration. ⺠AAVS1 locus of human chromosome 19 has been considered as a safe integration site for gene therapy. ⺠The plasmid-AAVS1 specific integration yields GLP-1 production in mice carrying AAVS1. ⺠This therapy improves metabolic status in the AAVS1 mice on high-fat diet feeding. ⺠This approach shows no detrimental effects and thus may find clinical applications.
