► The plasmodial surface anion channel exhibits voltage-dependent inactivation. ► Both inactivation and recovery are voltage-dependent with a small effective valence. ► Inactivation requires washout of cytosol and is affected by intracellular protease. ► A model based on charged domains of the channel protein is presented.
