► Neil3 is the major enzyme in tissues removing hydantoins from single stranded DNA. ► Neurospheres derived from newborn Neil3−/− forebrain display reduced proliferation but normal differentiation capability. ► Neil3−/− derived MEFs exhibit reduced proliferation and are cisplatin sensitive.
