► CaMy1 directly interacts with human brain SCG10 in Ca2+-dependent manner in vivo. ► CaMy1/SCG10 binding involves CaMy1 C-terminal domain and N-terminal SCG10 fragment. ► CaMy1 and SCG10 protein levels increase during early neural development. ► CaMy1 co-localizes with SCG10 in developing neurons. ► CaMy1 modulates SCG10-related neurite outgrowth and microtubule dynamics.
