Abstract
► We show DNA methylation dynamics in the CYP3A4 regulatory region in human livers. ► CpGs near CYP3A4 transcription factor (TF) binding regions show variable methylation. ► Single CpG methylation near proximal promoter and CLEM4 may predict CYP3A4 expression. ► Hypomethylation near TF binding regions may explain fetal/adult CYP3A4 shift. ► DNA promoter methylation dynamics may be used to explain variable CYP3A4 expression.
