Abstract
► Cell-free protein synthesis based on E. coli is limited by mRNA secondary structures. ► RNA chaperones and helicases cannot increase yields in E. coli cell-free systems. ► A miRNA hybridizing with the 5′ UTR of mRNAs enhances translation. ► miRNAs might provide a general mechanism to increase yields in cell-free systems.
