Abstract

The present work is focused on the structural and dynamical properties of dUTPase-inhibitor candidate and dUTPase/disaccharide mixtures investigated by Photon Correlation Spectroscopy (PCS) and Elastic Neutron Scattering (ENS).PCS findings emphasise that the dUTPase hydrodynamic radius in solution does not change at low disaccharide concentrations, while reveal, at high disaccharide concentration, a breakdown of the Stokes–Einstein law. On the other hand, the ENS findings show that the local mean square displacement of hydrated dUTpase/disaccharide systems is linearly dependent on the disaccharide/water viscosity, highlighting that the protein dynamics is coupled with that of the surrounding matrix.
