► Acute endotoxemia causes macrophage accumulation of in the lung and liver. ► Liver macrophages are more sensitive to acute endotoxemia than lung macrophages. ► The effects of acute endotoxemia are blunted in mice with a nonfunctional TLR4. ► TLR4 is key in oxidative stress, inflammation and macrophage activation.
